Abstract: 

复旦大学赵冰等 | 人胆道类器官的简易悬浮培养方案

本Technical Note聚焦胆道类器官的新型培养方式——悬浮培养。类器官是细胞在体外自组装或保留源结构形成的3D培养物。类器官较好地保留源结构的特性与功能,正成为生物基础研究、医学乃至环境生物学等多领域的热点。胆道类器官是来源于胆道的类器官。胆道类器官是研究肝胆疾病的良好模型,同时小鼠和离体人源肝脏模型均显示:胆道类器官能再生部分胆道,具有再生潜能。此外,胆管细胞可转化生成肝实质细胞;因此胆道类器官有望作为肝实质细胞来源候选库,应用于肝实质细胞疗法,缓解临床肝脏移植供体短缺的压力。胆道类器官的各项应用需要类器官培养量的支持。常规培养胆道类器官的方式是将类器官包埋于基质胶内,再覆没培养基,即静态培养。静态培养难以实现规模化,且操作相对冗杂。在本次研究中,我们将基质胶与培养基混合,形成基质胶悬液,构建了胆道类器官的悬浮培养方案。结果显示,悬浮培养能维持两种胆道类器官(成人胆管类器官HBD和人胚胆囊类器官FGO)的正常增殖,保留特性,在增殖能力和培养容量上更具优势。该方案有望实现胆道类器官的规模化培养,对接胆道类器官的多样应用前景。
