基因3型猪戊型肝炎病毒浙江株全基因组序列分析创新点:首次分析报道浙江地区基因3型猪戊型肝炎病毒全序列。 方法:利用套式PCR和末端快速扩增法对分离自浙江地区的戊型肝炎病毒进行分段扩增,经克隆测序后拼接全基因组。进一步利用生物信息学方法对基因组结构、开放阅读框及其基因型等进行分析,并与国内外戊型肝炎毒株进行比较分析。 结论:浙江株戊型肝炎病毒(ZhJ-PJ050-3)基因组由7223个核苷酸组成,其中3’非编码区含有23个碱基的poly(A)尾。全基因组包含ORF1、ORF2和ORF3三个主要基因,长度分别为5109、1983和342 bp。进化分析显示,ZhJ-PJ050-3为基因3型,与国内唯一报道的上海株3型毒株以及多数日本毒株共属3b亚型。与国内外戊型肝炎序列比对发现,ZhJ-PJ050-3基因组共有24个特有的点突变,且在ORF1中含有一个亮氨酸缺失。这些分子特征将为下一步的研究提供方向。 关键词组: Darkslateblue:Affiliate; Royal Blue:Author; Turquoise:Article
